Drug intelligence / Profile preview
Orynotide (also known as ORTD-1 or ORYN-1001) is a novel engineered macrocyclic peptide therapeutic developed by Oryn Therapeutics. Bioinspired by theta-defensins (cyclic host-defense peptides), orynotide acts as an immunomodulator that targets complex protein surfaces and allosteric regulatory domains. Its primary mechanism of action involves the inhibition of tumor necrosis factor-alpha convertase (TACE/ADAM17) and the subsequent modulation of downstream inflammatory pathways, including the NF-kappaB pathway. Orynotide is designed to reprogram dysregulated immune networks rather than broadly suppressing them, and has been investigated in clinical trials for the treatment of rheumatoid arthritis and COVID-19-related pneumonia.
