OS2966 is a first-in-class, humanized and deimmunized monoclonal antibody that targets CD29 (integrin β1), a cell surface adhesion receptor implicated in cancer cell growth, proliferation, invasion, angiogenesis, immune response modulation, and therapeutic resistance. By blocking the β1 integrin subunit (CD29), OS2966 disrupts multiple mechanisms driving malignancy. It is being developed primarily for high-grade glioma/glioblastoma and has received orphan drug designation from the FDA for these indications. The drug is delivered directly to the tumor site via convection-enhanced delivery (CED) to bypass the blood-brain barrier and achieve effective concentrations in central nervous system tumors. Preclinical studies have shown antiproliferative, anti-invasive, antivascularization, proapoptotic effects with significant tumor suppression or regression in models of glioblastoma[3][4][5][7].
