Overview

OS4T-eIL-12 is a **lipid nanoparticle (LNP) formulation containing engineered interleukin-12 (eIL-12) mRNA**, designed for systemic delivery to treat glioblastoma (GBM)[1]. The drug leverages the OS4T lipid nanoparticle platform to facilitate targeted mRNA delivery to brain tissue, inducing expression of IL-12 in situ. The mechanism leads to increased local immune activation, the upregulation of M1 macrophage/microglia markers (CD80, CD86, iNOS), enhanced activation of CD8^+^ T cells, and reduced regulatory CD4^+^ T cells, resulting in significant tumor growth inhibition and extended survival in preclinical models[1]. OS4T-eIL-12 is being investigated primarily for GBM in animal studies; there is no evidence from current results of clinical trials in humans.
