Overview

Osavampator is an investigational small molecule drug that acts as a positive allosteric modulator (PAM) of the alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA) receptor, a subtype of glutamate receptor. By enhancing AMPA receptor activity, osavampator increases glutamatergic neurotransmission and synaptic plasticity—mechanisms believed to underlie its antidepressant effects. Unlike traditional monoaminergic antidepressants or NMDA antagonists like ketamine, osavampator offers a targeted approach with minimal risk of dissociation or sedation. It is being developed primarily as an adjunctive therapy for major depressive disorder (MDD), especially in patients with inadequate response to standard treatments. The drug was originally discovered by Takeda and is now being developed globally by Neurocrine Biosciences (except in Japan, where Takeda retains rights). Osavampator has shown promising efficacy and safety results in Phase 2 trials and entered Phase 3 development in early 2025[1][3][4][5][6][7][8][9].
