A combination therapy of **osimertinib**, **pemetrexed**, and **carboplatin** is used primarily for the treatment of advanced non-small-cell lung cancer (NSCLC) harboring epidermal growth factor receptor (EGFR) mutations. **Osimertinib** is a third-generation EGFR tyrosine kinase inhibitor, selectively targeting EGFR-sensitizing and T790M resistance mutations. **Pemetrexed** is an antifolate antineoplastic agent that inhibits enzymes in folate metabolism, thereby disrupting DNA and RNA synthesis in rapidly dividing tumor cells. **Carboplatin** is a platinum-based chemotherapy compound that induces DNA cross-linking, causing apoptosis in cancer cells. This combination has shown improved progression-free survival and objective response rates compared to osimertinib alone, with a manageable safety profile, although increased chemotherapy-related adverse events occur. The combination is being investigated and increasingly used as first-line therapy, especially in patients with central nervous system involvement[1][3][5][7].
