Overview

Osmium complex 2 refers to a specific organometallic osmium(II) arene compound, most notably the lead candidates developed by the Sadler group at the University of Warwick. One prominent version is **FY26** ([Os(η6-p-cymene)(1,2-diaminoethane)I]PF6), while another is the catalytic complex **[Os(η6-p-cymene)(azopyridine-NMe2)I]PF6**. These complexes are designed as next-generation anticancer agents to overcome cisplatin resistance. Their mechanism of action typically involves a combination of DNA binding (often via intercalation or ligand-based interactions rather than direct coordination like cisplatin), the generation of reactive oxygen species (ROS), and the catalytic depletion of intracellular glutathione (GSH). By disrupting the redox balance of cancer cells and inducing mitochondrial dysfunction, these complexes exhibit high potency (often 50-100 times that of cisplatin) across various cancer cell lines, including those with acquired resistance to platinum-based therapies.
