Osmundacetone (OSC) is a bioactive phenolic compound isolated from the medicinal mushroom *Phellinus igniarius*. It exhibits potent antiproliferative and cytotoxic effects against non-small cell lung cancer (NSCLC) cells. The drug's mechanism of action centers on the downregulation of glutamate dehydrogenase 1 (GLUD1), a key enzyme in glutamine metabolism. By inhibiting the glutamine/glutamate/α-ketoglutarate (α-KG) metabolic axis, osmundacetone disrupts mitochondrial oxidative phosphorylation (OXPHOS) and reduces ATP production. This metabolic modulation leads to G2/M cell cycle arrest and suppression of tumor growth, as demonstrated in both in vitro and in vivo xenograft models.
