Drug intelligence / Profile preview
OspA-MATD is a preclinical two-component protein conjugate vaccine candidate being developed by Lochhead Scientific, in collaboration with the Medical College of Wisconsin (MCW), for the prevention of Lyme disease (caused by *Borrelia burgdorferi*). The vaccine consists of the outer surface protein A (OspA) of *Borrelia burgdorferi* as a B-cell immunogen covalently linked to the methyl-accepting transducer domain (MATD) of the *Borrelia burgdorferi* chemotaxis protein Mcp4, which serves as an immunodominant CD4+ T-cell antigen. By combining both B-cell and T-cell immunogens, OspA-MATD is designed to provide additional T-cell help, stimulating strong T-cell-dependent germinal center responses to OspA. This is intended to promote robust affinity maturation, isotype switching, and the generation of long-lived plasma cells, ultimately producing high-affinity, long-lasting antibody responses to OspA to overcome the short-term protection and frequent booster requirements of traditional OspA-only vaccines.
