The OspA-MATD conjugate vaccine is a preclinical-stage vaccine candidate for the prevention of Lyme disease, developed by Lochhead Scientific in partnership with the Medical College of Wisconsin. The vaccine utilizes a conjugate design that pairs lipidated *Borrelia burgdorferi* outer surface protein A (OspA) as a B-cell immunogen with the methyl-accepting transducer domain (MATD) of the chemotaxis protein Mcp4 as a T-cell immunogen. This approach is intended to elicit high-affinity, long-lasting protective antibodies by stimulating robust T cell-dependent germinal center responses, addressing the durability issues observed in earlier OspA-based vaccines. The Mcp4 MATD component was identified as an immunodominant CD4+ T cell antigen through immunopeptidomics, providing the necessary T-cell help to enhance the immune response against the *Borrelia burgdorferi* pathogen.
