Osperotug (BMS-986442) is an investigational, high-affinity, Fc-enhanced monoclonal antibody targeting the T-cell immunoreceptor with Ig and ITIM domains (TIGIT). TIGIT is an immune checkpoint receptor expressed on T cells and Natural Killer (NK) cells that inhibits their activation upon binding to ligands such as CD155. By blocking the TIGIT pathway, osperotug aims to restore and enhance the antitumor immune response. Developed by Bristol Myers Squibb, it is being evaluated primarily in combination with nivolumab (a PD-1 inhibitor) and chemotherapy for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC). The Fc-enhancement is designed to promote antibody-dependent cellular cytotoxicity (ADCC) or other Fc-mediated effector functions to further deplete TIGIT-expressing regulatory T cells within the tumor microenvironment.
