Overview

This synthetic oligopeptide, with the sequence GLRSKSKKFRRPDIQYPDATDEDITSHM, is derived from the collagen-binding motif (CBM) of the protein osteopontin (SPP1). It is being developed through a collaboration between Yonsei University College of Medicine and Nano Intelligent Biomedical Engineering Corporation (NIBEC) for the treatment of bone loss, with a specific focus on individuals with cerebral palsy. The peptide functions by binding directly to collagen and promoting osteoblast differentiation and bone mineralization. Its mechanism of action involves the activation of the Ca2+/calmodulin-dependent protein kinase II (CaMKII)/ERK/AP-1 signaling pathway. Preclinical evidence in mouse and rabbit models indicates that the peptide can increase trabecular thickness and upregulate bone formation markers such as Runx2 and osteocalcin, effectively reversing bone loss.
