Overview

Osteopontin peptide 2 is an experimental peptide-based vaccine candidate designed to target the pro-inflammatory activity of osteopontin (OPN), a cytokine involved in chronic low-grade inflammation associated with obesity and cardiovascular disease. The peptide sequence corresponds to the cryptic integrin-binding site (SLAYGLR in mice or SVVYGLR in humans) that is exposed only after OPN is proteolytically cleaved by thrombin. By inducing an active immune response against this specific motif, the vaccine was intended to neutralize the activity of cleaved OPN, thereby reducing adipose tissue inflammation, insulin resistance, and atherosclerosis. However, preclinical studies in murine models (Ldlr-/- and wild-type mice) demonstrated a lack of therapeutic efficacy. The failure was attributed to the vaccine's inability to induce antibodies that could effectively bind the target epitopes within the context of the full-length, functional OPN protein.
