Osteoprotegerin (OPG) is a cytokine receptor of the tumor necrosis factor (TNF) receptor superfamily that functions as a decoy receptor for RANKL (receptor activator of nuclear factor kappa-B ligand). It inhibits osteoclast formation and activity, thereby blocking bone resorption. OPG also binds to TRAIL (TNF-related apoptosis-inducing ligand) and inhibits TRAIL-induced apoptosis. It plays critical roles in bone metabolism regulation and has been investigated as a potential therapeutic for osteoporosis, cancer-induced bone disease, hypercalcemia of malignancy, and vascular disorders.
