Drug intelligence / Profile preview
OSUAB-0284 is a novel, small-molecule bacterial topoisomerase inhibitor (NBTI) under preclinical development for the treatment of multidrug-resistant bacterial infections, particularly methicillin-resistant Staphylococcus aureus (MRSA) and Bacillus anthracis (anthrax). Developed by researchers at The Ohio State University, OSUAB-0284 acts by dual-targeting bacterial type II topoisomerases, specifically DNA gyrase and topoisomerase IV. It inhibits supercoiling by DNA gyrase and decatenation by topoisomerase IV with balanced potency, promoting the accumulation of single-strand DNA breaks. Due to its unique binding mechanism, OSUAB-0284 maintains activity against fluoroquinolone-resistant strains, making it a promising candidate for addressing antimicrobial resistance.
