Drug intelligence / Profile preview
OSURALi is a novel small molecule allosteric inhibitor of the small GTPases Ras-Like Oncoproteins A and B (RALA and RALB). Developed by researchers at The Ohio State University, OSURALi binds to the allosteric pocket of RALA and RALB, potently suppressing their activation (GTP binding). Unlike earlier-generation RAL inhibitors such as RBC8 and BQU57, which exhibit off-target toxicities, OSURALi demonstrates selective cytotoxicity in RAL-dependent triple-negative breast cancer (TNBC) cell lines while sparing normal and RAL-independent cells. In preclinical models of TNBC, OSURALi is well tolerated and significantly reduces primary tumor growth and spontaneous lung metastasis.
