OT-003 is a proprietary small molecule modulator of the Tropomyosin receptor kinase B (TrkB), developed by Pangea Bio (Pangea Botanica Ltd). It is designed to promote neuronal survival and plasticity, reduce pathological protein aggregation, and alleviate functional disability in central nervous system (CNS) diseases. Preclinical studies have shown that OT-003 reduces dopaminergic neuronal cell death, neurite network loss, mitochondrial stress, and alpha-synuclein aggregation in models of Parkinson’s disease. The compound activates TrkB signaling pathways, which are implicated in enhancing neuronal survival, synaptic plasticity, reducing neuroinflammation and pathological protein aggregates such as α-synuclein, amyloid-β and tau. These effects lead to improvements in memory, cognition, and motor function as demonstrated in animal models. As of mid-2025, OT-003 is undergoing clinical-enabling safety pharmacology and toxicology studies with plans for Phase 1 clinical trials to begin by Q1 2025[1][2][3].
