OT-1301 is a sustained-release intraocular or intra-corneal implant containing cyclosporin, a calcineurin inhibitor, developed by Ocumension Therapeutics. It is specifically designed to provide localized immunosuppression to prevent corneal graft rejection following transplantation. By inhibiting calcineurin, the drug prevents the dephosphorylation of the nuclear factor of activated T-cells (NFAT), thereby blocking the transcription of interleukin-2 (IL-2) and other cytokines essential for T-cell activation and proliferation. This localized delivery system aims to maintain therapeutic drug levels at the graft site while minimizing the systemic side effects and compliance issues associated with topical or systemic immunosuppressants.
