OT-82 is a novel, orally active small molecule inhibitor of nicotinamide phosphoribosyltransferase (NAMPT), an enzyme critical for the NAD salvage pathway. By inhibiting NAMPT, OT-82 depletes cellular NAD levels and inhibits NAD-dependent enzymes necessary for rapid cell proliferation, leading to selective cell death in NAMPT-overexpressing cancer cells. Preclinical studies have shown strong efficacy against hematopoietic malignancies such as acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), erythroleukemia, multiple myeloma, and Burkitt's lymphoma. The drug has demonstrated selectivity for malignant hematopoietic cells over healthy bone marrow precursors and a favorable toxicity profile compared to other NAMPT inhibitors. Developed by OncoTartis, its clinical development is focused on blood cancers including relapsed or refractory lymphoma[1][3][5][6].
