Overview

OT-C001 + rituximab + interleukin-2 is a combination drug regimen, experimentally investigated primarily for the treatment of non-Hodgkin lymphoma (NHL). The combination includes OT-C001 (details about OT-C001 are not available in current authoritative sources and may represent a code name for a specific investigational or proprietary biologic or compound), rituximab (a chimeric monoclonal antibody targeting CD20 on B lymphocytes), and interleukin-2 (IL-2), a recombinant cytokine that promotes proliferation and activation of lymphocytes such as NK cells and T-cells. Mechanistically, rituximab induces antibody-dependent cellular cytotoxicity (ADCC) and apoptosis of CD20+ B-cells, while interleukin-2 enhances expansion and cytotoxic activity of natural killer (NK) and other immune effector cells, potentially increasing rituximab's efficacy. This combination has been tested in phase 2 trials for various B-cell lymphomas, mostly in refractory or previously treated patients, but combination efficacy (especially with just rituximab plus IL-2 and not specifically OT-C001 + rituximab + IL-2) has shown limited clinical benefit in certain populations, despite immune cell expansion and increased ADCC in vitro[1][2][3][4][5][6][7].
