OT1 TCR-engineered T cells (also referred to as OT1-iT cells) are a specialized preclinical research construct used to model adoptive T-cell therapy in murine systems. These T cells are genetically engineered, typically via retroviral or lentiviral transduction, to express the OT1 T-cell receptor (TCR). The OT1 TCR is a mouse MHC class I-restricted receptor that specifically recognizes the SIINFEKL peptide (residues 257–264) of the ovalbumin (OVA) protein when presented in the context of the H2-Kb molecule. This system is widely utilized in mouse models to study antigen-specific T-cell responses, including activation, expansion, and anti-tumor efficacy against solid tumors (such as B16 melanoma or EL4 lymphoma) that have been engineered to express ovalbumin as a surrogate tumor-associated antigen (TAA).
