OTD70DERM is a topical gel formulation of dapansutrile (also known as OLT1177), a selective small-molecule inhibitor of the NLRP3 (NACHT, LRR, and PYD domains-containing protein 3) inflammasome. Developed by Olatec Therapeutics, it is primarily being investigated for the treatment of atopic dermatitis and other inflammatory skin conditions. The drug's mechanism involves binding to the NLRP3 protein, which prevents the assembly of the inflammasome complex and the subsequent activation of caspase-1. This inhibition blocks the maturation and secretion of the pro-inflammatory cytokines interleukin-1β (IL-1β) and interleukin-18 (IL-18). By targeting these specific mediators of the innate immune response, OTD70DERM aims to reduce skin inflammation and pruritus without the broad immunosuppressive effects associated with corticosteroids or calcineurin inhibitors.
