Overview

OTI-900 is a first-in-class small molecule inhibitor of Chromodomain-helicase-DNA-binding protein 1-like (CHD1L), also known as ALC1, currently in preclinical development by Onconaut Therapeutics. CHD1L is a macrodomain-containing DNA-dependent ATPase and chromatin remodeler that is frequently overexpressed or amplified in several solid tumors, including hepatocellular carcinoma, lung, and breast cancers. It plays a pivotal role in the DNA damage response and chromatin relaxation. OTI-900 is designed to selectively inhibit the activity of CHD1L, thereby disrupting DNA repair mechanisms and inducing synthetic lethality or growth arrest in cancer cells that depend on this oncogene for survival.
