Otlertuzumab (TRU-016) is a humanized anti-CD37 protein therapeutic that induces direct caspase-independent apoptosis of malignant B cells and triggers antibody-dependent cell-mediated cytotoxicity. It is designed to target CD37, a tetraspanin expressed on normal and malignant B cells. Bendamustine is an alkylating agent of the nitrogen mustard class that forms DNA crosslinks leading to cell death; it is used in the treatment of chronic lymphocytic leukemia (CLL) and indolent B-cell non-Hodgkin lymphoma (NHL). The combination has been studied for relapsed or refractory CLL and indolent NHL[1][3][5].
