Overview

OTO-5XX is a small molecule drug program developed by Otonomy, Inc., in collaboration with the University of California, aimed at preventing cisplatin-induced hearing loss (CIHL). Classified as a neurotransmitter stimulant, the drug is designed to be administered otically (into the ear) to provide local otoprotection. A key feature of the program is its ability to protect the inner ear from the ototoxic effects of cisplatin without providing 'tumor protection,' thereby ensuring that the chemotherapy's systemic efficacy against cancer remains uncompromised. As of 2018, the program was in the preclinical stage, with efforts focused on clinical candidate selection and IND-enabling studies, particularly targeting the unmet need in pediatric oncology patients.
