OTS-412 is an oncolytic vaccinia virus (OVV) developed by Bionoxx for the treatment of solid tumors. It is engineered from a Wyeth strain vaccinia virus backbone with a deletion of the native thymidine kinase (VV-tk) gene and the insertion of a mutant herpes simplex virus thymidine kinase (HSV-tk) transgene. This modification enhances tumor selectivity and provides a safety mechanism, as the HSV-tk gene allows viral replication to be controlled or terminated via the administration of the antiviral prodrug ganciclovir (GCV). OTS-412 is designed to induce direct oncolysis and stimulate a systemic anti-tumor immune response. A modified version, mOTS-412, has received FDA IND clearance for Phase 1 clinical trials in patients with immune checkpoint inhibitor-resistant solid tumors, both as a monotherapy and in combination with anti-PD-L1 therapy.
