Overview

OTX-DED is an investigational, physician-administered, preservative-free intracanalicular insert designed for the short-term treatment of the signs and symptoms of dry eye disease. It delivers a sustained and tapered dose of dexamethasone (0.2 or 0.3 mg) directly to the ocular surface over approximately 2–3 weeks via a biodegradable polyethylene glycol hydrogel matrix placed in the canaliculus. The mechanism combines anti-inflammatory therapy (via corticosteroid action) with tear conservation through punctal occlusion, offering an alternative to conventional steroid eye drops and potentially reducing side effects associated with preservatives in topical formulations[1][3][4][6]. Developed by Ocular Therapeutix, it is currently in Phase 2 clinical trials for dry eye disease.
