OTX-IVT is an investigational, sustained‑release intravitreal formulation of the anti‑VEGF biologic aflibercept, developed using Ocular Therapeutix’s bioresorbable hydrogel-based drug delivery technology in collaboration with Regeneron. It is designed as an extended‑delivery depot system to provide controlled, long‑acting intravitreal release of aflibercept over several months, with the goal of reducing injection frequency while maintaining VEGF inhibition for retinal vascular diseases such as diabetic macular edema and other ophthalmic indications.[10][20] Preclinical work with intravitreally injected aflibercept-DDS has shown controlled release of bioactive aflibercept in primate eyes for up to 6 months without adverse effects, supporting its potential as a long-acting anti‑VEGF treatment platform.[10]
