Overview

OURO-117 is a selective allosteric inhibitor of tyrosine kinase 2 (TYK2) currently in Phase 2 clinical development for the treatment of various inflammatory diseases. Developed using Ouro Medicines' ATAC kinase targeting platform, the drug was acquired by Gilead Sciences in a $3.2 billion acquisition to bolster their oral immunology and inflammation pipeline. OURO-117 functions by binding to the TYK2 pseudokinase domain, which induces a conformational change that inhibits the catalytic activity of the adjacent kinase domain. This allosteric approach provides high selectivity for TYK2 over other Janus kinase (JAK) family members, potentially reducing the risk of off-target toxicities such as cytopenia or lipid elevations often associated with non-selective JAK inhibition.
