OV1203 is a selective small molecule inhibitor of Rho-associated coiled-coil containing protein kinase 2 (ROCK2), currently in development by Ovid Therapeutics in collaboration with Graviton Bioscience. It is designed as a second-generation candidate within Ovid's ROCK2 inhibitor portfolio, specifically optimized for the treatment of Cerebral Cavernous Malformations (CCM) and potentially other neurovascular conditions. The mechanism of action involves the inhibition of the ROCK2 signaling pathway, which is a critical regulator of the cellular cytoskeleton, endothelial cell adhesion, and vascular permeability. In CCM, the loss of vascular integrity leads to the formation of fragile, leaky capillary lesions in the central nervous system; OV1203 aims to restore endothelial barrier function and prevent the growth or hemorrhage of these lesions. The drug is part of a broader library of ROCK2 inhibitors licensed by Ovid to address rare neurological diseases with high unmet need.
