Overview

OV350 is a first-in-class small molecule drug that directly activates the potassium chloride cotransporter isoform 2 (KCC2), a neuronal membrane protein critical for maintaining chloride homeostasis and regulating neuronal excitability. By activating KCC2, OV350 enhances inhibitory neurotransmission and reduces neuronal hyperexcitability, which is implicated in epilepsy and other central nervous system disorders. Preclinical studies have demonstrated that OV350 can terminate benzodiazepine-resistant seizures, improve survival following nerve agent-induced status epilepticus, restore the efficacy of diazepam in resistant models, and provide neuroprotective effects by limiting seizure-associated brain injury. The compound was originally developed by AstraZeneca and later licensed to Ovid Therapeutics, which is currently advancing its clinical development primarily for treatment-resistant epilepsies.
