OVA-mRNA dendritic cell vaccine is an experimental cancer immunotherapy consisting of dendritic cells (DCs) that have been ex vivo electroporated with mRNA encoding the model antigen ovalbumin (OVA). This vaccine is primarily used in preclinical research to study DC migration, antigen presentation, and T-cell activation in tumor models such as GL261-OVA glioblastoma. The mechanism involves the translation of the mRNA within the DCs, followed by the processing and presentation of OVA-derived peptides on MHC molecules to prime and expand antigen-specific CD8+ and CD4+ T cells. In research conducted at Duke University, it is often studied alongside adjuvants like CCL3 or tetanus-diphtheria (Td) preconditioning to enhance DC migration to draining lymph nodes and improve overall survival in established tumor models.
