OVV-αCD47nb is an engineered oncolytic vaccinia virus (OVV) designed to express and secrete an anti-CD47 nanobody (αCD47nb) directly within the tumor microenvironment. Developed by Hangzhou Ronggu Biotechnology in collaboration with academic institutions like Zhejiang University and Hangzhou Medical College, this viro-immunotherapy combines direct viral oncolysis with localized immune checkpoint blockade. By targeting CD47, the secreted nanobody blocks the "don't eat me" signal (CD47-SIRPα interaction), thereby enhancing macrophage-mediated phagocytosis of tumor cells. Preclinical studies in breast cancer, colon cancer, pancreatic cancer, and multiple myeloma models have demonstrated that intratumoral administration of OVV-αCD47nb promotes immune cell infiltration, activates CD8+ T cells, polarizes tumor-associated macrophages toward an antitumoral M1 phenotype, and synergizes with PD-1 inhibitors or proteasome inhibitors like bortezomib.
