OVV-OPCML is an oncolytic virus therapy consisting of a recombinant vaccinia virus (Copenhagen strain) engineered to express the Opioid Binding Protein/Cell Adhesion Molecule-Like (OPCML) tumor suppressor gene. OPCML is a GPI-anchored protein frequently inactivated by somatic methylation in epithelial ovarian cancer (EOC). The drug exerts its therapeutic effect through multiple mechanisms: direct viral-mediated oncolysis of tumor cells, restoration of OPCML protein expression which inhibits receptor tyrosine kinases (RTKs) such as EGFR, ERBB2, and FGFR1, and suppression of tumor angiogenesis via downregulation of VEGFR2 signaling. Additionally, OVV-OPCML promotes an anti-tumor immune response by increasing CD8+ T cell infiltration and elevating levels of IFN-γ and TNF-α, showing synergistic potential when combined with PD-1 checkpoint inhibitors.
