Overview

OX-40 ligand (OX40L), also known as CD252 or TNFSF4 (tumor necrosis factor ligand superfamily member 4), is a type II transmembrane glycoprotein and an endogenous protein in humans. It is the natural ligand for the receptor OX40 (CD134/TNFRSF4), which is expressed on activated T cells. OX40L itself is stably expressed on antigen-presenting cells such as dendritic cells (especially DC2s), macrophages, activated B lymphocytes, and some non-immune cells like endothelial and smooth muscle cells[1][2][5]. The interaction between OX40 and its ligand delivers a late co-stimulatory signal that promotes T cell activation, survival, proliferation of activated CD4+ T cells, development of memory T cells and polarization towards Th2 immune responses[1][5][7]. This pathway plays a critical role in adaptive immunity. Modulation of the OX40/OX40L axis has been explored for therapeutic purposes in cancer immunotherapy as well as autoimmune/inflammatory diseases[2][3][7].
