OX382 is an investigational, potent, and highly selective oral orexin-2 receptor (OX2R) agonist developed by Centessa Pharmaceuticals through its subsidiary Orexia Therapeutics. Utilizing structure-based drug design and the StaR (Stabilized Receptor) technology, OX382 is engineered to mimic the activity of the endogenous neuropeptide orexin, which is crucial for regulating wakefulness and is notably deficient in patients with Narcolepsy Type 1 (NT1). By selectively activating OX2R, OX382 aims to address the underlying pathophysiology of narcolepsy and other sleep disorders characterized by excessive daytime sleepiness (EDS). It is designed with a pharmacokinetic profile intended to provide 24-hour coverage, potentially offering a differentiated treatment option for patients with chronic sleep-wake disorders. The molecule is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and pharmacokinetic profile.
