OX40 agonist VHH antibody is a preclinical-stage cancer immunotherapy being developed by COGNANO. It is a single-domain antibody (VHH), also known as a nanobody, designed to bind and activate the OX40 receptor (CD134), a co-stimulatory member of the tumor necrosis factor receptor superfamily (TNFRSF) found on activated T cells. By agonizing OX40, the antibody enhances T-cell proliferation, survival, and effector functions, such as cytokine release, while potentially reducing the immunosuppressive activity of regulatory T cells (Tregs). The VHH platform, derived from camelid heavy-chain-only antibodies, offers advantages in terms of stability, small size, and the ability to construct multivalent molecules, which can more effectively cluster OX40 receptors to trigger downstream signaling compared to traditional bivalent monoclonal antibodies.
