Overview

This combination regimen is a multi-modal therapy being evaluated by Peking University for the treatment of advanced hepatocellular carcinoma (HCC), specifically in patients with portal vein tumor thrombosis (PVTT). The regimen integrates local and systemic approaches: Hepatic Arterial Infusion Chemotherapy (HAIC) using the FOLFOX protocol (comprising oxaliplatin, 5-fluorouracil, and leucovorin) is administered directly into the hepatic artery to provide high local concentrations of cytotoxic agents. This is combined with systemic administration of lenvatinib, a multi-targeted tyrosine kinase inhibitor that blocks angiogenesis and tumor cell proliferation, and a PD-1 inhibitor, an immune checkpoint therapy that restores the anti-tumor activity of T-cells. The approach aims to achieve synergistic local tumor control and systemic disease management.
