Overview

Oxaliplatin + capecitabine + pembrolizumab is a combination regimen used primarily for the treatment of advanced or metastatic cancers, including oesophageal and gastro-oesophageal junction adenocarcinoma that express PD-L1, as well as being investigated in other solid tumors such as biliary tract cancer. Capecitabine and oxaliplatin are chemotherapy agents; capecitabine is an oral prodrug of 5-fluorouracil that inhibits DNA synthesis by interfering with thymidylate synthase, while oxaliplatin is a platinum-based agent causing DNA crosslinking and apoptosis. Pembrolizumab is a monoclonal antibody immune checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor on T cells, thereby enhancing anti-tumor immune responses. This combination leverages both cytotoxic chemotherapy to reduce tumor burden and immunotherapy to stimulate the patient’s immune system against cancer cells[1][2][3].
