Overview

FOD is a combination chemotherapy regimen comprising fluorouracil, oxaliplatin, and doxorubicin. It is primarily investigated as a neoadjuvant (preoperative) treatment for patients with locally advanced gastric cardia cancer, a specific type of esophageal or gastric adenocarcinoma. The regimen's efficacy is derived from the synergistic effects of its three components: fluorouracil, a pyrimidine analog antimetabolite that inhibits DNA synthesis; oxaliplatin, a third-generation platinum agent that creates DNA cross-links; and doxorubicin, an anthracycline that disrupts DNA repair and transcription by intercalating into DNA and inhibiting topoisomerase II. Research conducted by The First Affiliated Hospital of Zhengzhou University has focused on using this combination to downstage tumors and improve the rate of complete surgical resection (R0 resection) in patients with cardia-located gastric malignancies.
