oxaliplatin + S-1 + toripalimab

Oxaliplatin + S-1 + toripalimab is a chemoimmunotherapy combination regimen primarily investigated for the treatment of locally advanced, HER2-negative gastric or gastroesophageal junction adenocarcinoma. The regimen consists of three distinct agents: **oxaliplatin**, a platinum-based alkylating agent that induces DNA crosslinking; **S-1**, an oral fluoropyrimidine combination drug (comprising tegafur, gimeracil, and oteracil) that acts as a thymidylate synthase inhibitor; and **toripalimab** (brand name Loqtorzi), a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. This combination is designed to leverage the cytotoxic effects of chemotherapy to promote immunogenic cell death, thereby enhancing the anti-tumor T-cell activity restored by the PD-1 checkpoint inhibitor. It is currently being evaluated in the Phase II/III RESOLVE-2 trial (NCT03691454) as a neoadjuvant and adjuvant therapy.
