oxaliplatin + serplulimab

Oxaliplatin + serplulimab is a combination therapeutic regimen comprising the platinum-based chemotherapy agent oxaliplatin and the anti-PD-1 monoclonal antibody serplulimab (Hansizhuang). Serplulimab, developed by Shanghai Henlius Biotech, is designed to block the interaction between the PD-1 receptor on T cells and its ligands (PD-L1 and PD-L2), thereby reactivating the immune system's ability to recognize and destroy tumor cells. Oxaliplatin contributes to the regimen by inducing DNA damage through the formation of intra- and inter-strand cross-links, which triggers apoptosis in rapidly dividing cancer cells. This combination is primarily investigated for the treatment of gastric adenocarcinoma and gastroesophageal junction (GEJ) adenocarcinoma, often as part of the SOX (S-1 plus oxaliplatin) regimen. Key clinical evidence includes the Phase 3 ASTRUM-006 trial, which demonstrated significant improvements in event-free survival and pathological complete response rates when serplulimab was added to perioperative SOX chemotherapy for patients with PD-L1-positive resectable gastric/GEJ cancers.
