Overview

Oxatomide is a sedating antihistamine of the piperazine class, primarily used for the treatment and prevention of allergic symptoms such as allergic rhinitis, urticaria (hives), dermal pruritus, eczema, dermatitis, and prurigo. It acts as an antagonist at the histamine H1 receptor (HRH1), thereby inhibiting histamine-mediated effects including smooth muscle contraction and increased capillary permeability. In addition to its antihistaminic activity, oxatomide stabilizes mast cells—reducing degranulation and release of mediators like histamine, leukotrienes (LTC4), prostaglandin D2 (PGD2), and tryptase—and inhibits inflammatory cell recruitment and activation. These properties contribute to its anti-allergic and anti-inflammatory effects[1][2][3][4][5][6][7]. Oxatomide has also been investigated for use in asthma management due to these additional actions but is mainly approved for allergic conditions[7].
