Oxibendazole is a benzimidazole-class anthelmintic drug primarily used in veterinary medicine to treat and prevent infestations by roundworms, strongyles, threadworms, pinworms, and lungworm in horses and other domestic animals[2][4][5]. It acts as a small molecule polymerase inhibitor. The mechanism of action involves binding to the colchicine-sensitive site of β-tubulin in parasitic cells, inhibiting microtubule polymerization. This disruption impairs glucose uptake and depletes glycogen stores in the parasite, leading to energy depletion (decreased ATP production), paralysis, and death[1][6][7][8]. Oxibendazole is poorly absorbed orally; most remains in the gastrointestinal tract where it exerts its effect on gut-dwelling parasites[8]. It undergoes hepatic metabolism. While widely used for animals, it has also been investigated for human use against helminth intestinal infections (phase III trials)[1][3].
