Oxidized mannan-MUC1 (M-FP) is an investigational cancer immunotherapy vaccine developed by Prolipsis Medical Center in collaboration with researchers in Australia and Greece. It consists of the MUC1 fusion protein covalently conjugated to oxidized mannan. The vaccine's mechanism of action involves targeting the mannose receptor (CD206) on antigen-presenting cells, such as macrophages and dendritic cells, to facilitate the uptake and processing of the MUC1 antigen. This process elicits a robust cellular (Th1) and humoral immune response against MUC1-expressing tumor cells. MUC1 is a high molecular weight glycoprotein that is frequently overexpressed and aberrantly glycosylated in various adenocarcinomas, including breast cancer. In a randomized, double-blind Phase III pilot study, the vaccine demonstrated a statistically significant reduction in cancer recurrence and metastases in patients with Stage II breast cancer over a long-term follow-up period of 5.5 to 15 years.
