Overview

OXM8 is a synthetic oxyntomodulin (OXM) analogue developed as a peptide-based biologic for the potential treatment of obesity and weight management. Structurally, it is derived from the 29-amino acid sequence of glucagon, incorporating 5-7 single substitutions between residues 12 and 29 and an additional 2-3 amino acids at the C-terminus to improve metabolic stability. OXM8 acts as a dual agonist of the glucagon-like peptide-1 (GLP-1) receptor and the glucagon receptor. In preclinical rat models, daily administration of OXM8 resulted in a significant reduction in body weight (4-6% over 7 days); however, it paradoxically increased food intake, a finding attributed to its glucagon receptor activation profile.
