Oxybutynin, tolterodine, and solifenacin are three distinct small molecule pharmaceutical drugs, all belonging to the class of antimuscarinics (also known as anticholinergics), primarily indicated for the treatment of overactive bladder (OAB) symptoms, including urinary incontinence, urinary frequency, and urinary urgency. Each drug works by blocking muscarinic acetylcholine receptors, predominantly the M3 subtype, on the smooth muscle of the bladder. This action inhibits the involuntary contractions of the detrusor muscle, thereby relaxing the bladder, increasing its capacity, and reducing the urge to urinate. While they share a common mechanism, they differ in their selectivity for muscarinic receptor subtypes and their pharmacokinetic profiles, which can influence their side effect profiles and clinical use. Oxybutynin is available in oral and transdermal formulations, tolterodine in oral and transdermal gel forms, and solifenacin is available orally. These drugs are often compared in clinical practice for their efficacy and tolerability in managing OAB.
