Oxygen-ozone gas mixture is a medical gas therapy composed of a specific concentration of ozone (O3) in medical-grade oxygen (O2). Developed for clinical investigation by El Hospital Universitario De Gran Canaria Dr. Negrin, it is primarily being studied for the treatment of chemotherapy-induced peripheral neuropathy (CIPN). The mixture acts as a mild, controlled oxidative stressor that triggers the activation of the Nrf2 (nuclear factor erythroid 2-related factor 2) signaling pathway, leading to the upregulation of antioxidant enzymes such as superoxide dismutase and glutathione peroxidase. Additionally, it improves tissue oxygenation by increasing 2,3-diphosphoglycerate (2,3-DPG) levels in red blood cells, which shifts the hemoglobin-oxygen dissociation curve to the right, and modulates the release of anti-inflammatory cytokines. In patients with CIPN, these mechanisms are intended to reduce oxidative damage to peripheral nerves and improve microcirculatory blood flow, thereby alleviating neuropathic pain and sensory deficits.
