Overview

Oxyphenbutazone is a nonsteroidal anti-inflammatory drug (NSAID) and an active metabolite of phenylbutazone. It was primarily used for the symptomatic relief of musculoskeletal pain, arthritis (including ankylosing spondylitis, osteoarthritis, and rheumatoid arthritis), and gout. The drug acts mainly by inhibiting cyclooxygenase (COX) enzymes—both COX-1 and COX-2—thereby reducing prostaglandin synthesis involved in inflammation and pain signaling. Oxyphenbutazone was withdrawn from markets worldwide in the mid-1980s due to severe adverse effects such as bone marrow suppression, blood dyscrasias, and risk of Stevens–Johnson syndrome[2][3][4][5][6]. It has also been used topically for ocular inflammation abroad.
