OyaGen-Vif-HIV refers to a preclinical drug development program by OyaGen focused on identifying and optimizing small molecule inhibitors of the HIV-1 Viral infectivity factor (Vif). Vif is a critical viral accessory protein that HIV uses to evade the human innate immune system. Specifically, Vif recruits a host ubiquitin ligase complex to trigger the proteasomal degradation of APOBEC3G (A3G), a human enzyme that would otherwise inhibit viral replication. By inhibiting the Vif protein, OyaGen's lead compounds prevent the degradation of A3G, allowing it to be incorporated into budding HIV virions. Once inside the virion, A3G induces extensive C-to-U mutations in the viral genome during reverse transcription, effectively neutralizing the virus's ability to replicate and restoring the host's natural antiviral defense.
