Ozuriftamab vedotin is a conditionally and reversibly active antibody-drug conjugate (ADC) targeting receptor tyrosine kinase-like orphan receptor 2 (ROR2), a transmembrane protein overexpressed in various solid tumors, including head and neck squamous cell carcinoma, non-small cell lung cancer, triple-negative breast cancer, melanoma, ovarian cancer, and others. The drug consists of an anti-ROR2 monoclonal antibody linked to the cytotoxic agent monomethyl auristatin E. Its unique design allows selective binding to ROR2 in the acidic tumor microenvironment while minimizing binding in healthy tissues through a protein-associated chemical switch mechanism. This targeted approach aims to deliver potent antitumor activity with reduced off-tumor toxicity. Ozuriftamab vedotin is being developed by BioAtla for patients with advanced or refractory solid tumors who have progressed after standard therapies[1][2][3][5][6].
